"Protein Steroylation; Catalysis and Application"
Covalent modification of proteins by sterols represents one of the rarest forms of
post-translational modification. In the entire human proteome, with > 30,000 polypeptides, the total number of sterol-modified proteins stands at 3, and these three proteins make-up the so-called hedgehog family. Hedgehog proteins attract fundamental interest as cell-cell signaling factors during animal development, as well as oncogenic growth factors in multiple cancers. In both contexts, hedgehog function appears to depend on covalent steroylation.
In this talk, I will describe our recent efforts to probe the molecular mechanism of sterol modification, to manipulate the process with small molecules, and to apply the chemistry toward the emerging field of bioconjugation.
