Targeting Viral Surface Glycoproteins
Viral surface glycoproteins provide a powerful way to train the immune system to be prepared for a given virus. Elegant studies have demonstrated how stabilization of these metastable proteins can be undertaken. However, efforts were largely performed manually and step-by-step. We have incorporated several of these molecular strategies into systematic automation protocols to stabilize viral surface proteins while paying close attention to their molecular functionalities. Using our approach, we have generated a stabilized version for instance for fusion protein of RSV, rivaling the clinical candidate. We can provide further preliminary data on human metapneumovirus (hMPV) and SARS CoV-2 as model systems which both are otherwise highly instable. We will also shortly discuss potential therapeutic approaches for treatment of viral infections through targeting of intermediate states, as well as a new approach for the generation of protein-protein interactions using neural networks.
